Activity Description:
Targeted monoclonal antibodies have dramatically improved outcomes for patients with cancer by improving efficacy and reducing toxicities. Antibody-drug conjugates (ADCs) couple a monoclonal antibody for targeting with a cytotoxic therapy through a flexible linker that undergoes lysosomal degradation when internalized by cancer cells. While ADCs offer effective anti-cancer therapy for many patients, some are associated with meaningful, albeit manageable, ocular toxicities. For example, for patients with relapsed/refractory multiple myeloma, targeting B-cell maturation antigen (BCMA) has proven an effective anti-cancer approach, yet many experienced corneal toxicities in clinical trials, many of which could be managed. In this educational podcast series, experts in the fields of ophthalmology and oncology will review the mechanisms behind the ocular toxicities seen with ADC therapy, the frequency of these ocular toxicities and strategies to mitigate and manage them, and best practices to optimize the multidisciplinary team to reduce the burden of these adverse effects.

Upon successful completion of this activity, participants should be better able to:
• Summarize the latest understanding regarding the mechanisms underlying ocular toxicities associated with antibody-drug conjugates (ADCs) for cancer treatment.
• Describe the types and frequency of ocular toxicities associated with ADCs.
• Outline strategies for the mitigation and management of ocular toxicities associated with treatments for multiple myeloma.
• Summarize effective multidisciplinary strategies, including patient education, for the recognition, mitigation, and management of ocular toxicities associated with  BCMA-targeted therapy.
