A breast cancer diagnosis is now the most frequent cancer found around the globe. The Estrogen Receptor (ER) transcriptional pathway is responsible for the development of most breast cancers; recent discoveries regarding how the ER regulates gene expression have uncovered numerous therapeutic targets. The core treatment strategies for ER-positive breast cancers are endocrine therapies, which either disrupt the transactivation of ER or reduce the amount of ER present intracellularly. However, intrinsic and acquired resistance limits the clinical efficacy of these therapies, making them less effective overall.
A scientific understanding of the various subtypes of breast cancer has not only offered exceptional insight into the disease's inherent heterogeneity. Still, it has also shown critical processes related to the course of the disease and the patient's response to treatment. These relatively recent developments in our understanding of breast cancer subtypes have led to the creation of individualized treatment plans for distinct patient subpopulations, which has led to an increase in the overall survival rate.
The treatment of breast cancer has been revolutionized due to the emergence of more recent therapies that target the ER pathway (for example, CDK4/6 inhibitors that target a downstream target gene of the ER pathway). Recent research into the interaction between the estrogen receptor (ER) and other nuclear receptor transcription factors, such as the androgen receptor (AR), the progesterone receptor (PR), and the glucocorticoid receptor (GR), has led to the discovery of novel therapeutic opportunities for inhibiting ER activity through the activation of parallel nuclear receptor pathway competitions.
The treatment responses of individuals with HER2+ disease have significantly improved due to more recent breakthroughs in HER2-targeted medicines for the treatment of HER2+ malignancies. Our understanding of Triple Negative Breast Cancer (TNBC) has generally lagged behind our understanding of other subtypes of the disease; however, significant recent new insights into the genetic perturbations and the associations with specific treatment responses have shed light on this subtype of the disease.
Our understanding of breast cancer has been fundamentally altered by genomic and transcriptomic research, which has inspired the development of novel diagnostic and treatment approaches. Our understanding of breast cancers has been revolutionized by more recent advancements in single-cell RNA sequencing (scRNA-seq) and spatial imaging technologies, which have provided insight into complex cellular ecosystems that continuously interact to govern tumor progression and treatment outcome. These technologies have also provided new research opportunities. New computational approaches have made the development of spatial maps of breast cancers at single-cell resolution possible. These maps include immune cell populations and their interactions with stromal and tumor cells. These maps provide unique biological and mechanistic insights into the tumor microenvironment and its role in disease progression, treatment response, and resistance.
Experts in the field who investigate the biological basis of breast cancer, devise methods for patient stratification, conduct clinical trials, or create new treatments for patients with breast cancer will participate in this meeting.
