Tyrosine kinase inhibitors (TKIs) have recently demonstrated promising efficacy and safety outcomes in clinical trials of patients with retinal diseases, including neovascular age-related macular degeneration (nAMD) or diabetic retinopathy (DR). These novel agents may provide an alternative to the current standard of care—frequent injections of an anti-vascular endothelial growth factor (VEGF) agent, which is associated with substantial effects on patient comfort and treatment adherence. Current clinical trials are evaluating multiple TKIs delivered via delayed-release implant or suprachoroidal suspension, which may reduce patient burden and increase patient adherence to the treatment regimen. In this monograph, expert faculty review the challenges and limitations of the current methods of retinal disease management, detail the mechanisms by which TKIs affect retinal disease progression, report the latest clinical evidence on emerging TKIs for patients with nAMD and DR, and discuss the role and impact of TKIs on existing treatment paradigms.
