OVERVIEW

Neurocutaneous syndromes, also referred to as phakomatoses, are a group of rare but clinically significant disorders that affect both the nervous system and the skin. This CME/CE course provides a comprehensive review of the diagnosis, clinical features, and multidisciplinary management of major neurocutaneous syndromes, including Tuberous Sclerosis Complex (TSC), Neurofibromatosis Types 1 and 2 (NF1, NF2), and Sturge-Weber Syndrome (SWS).

Participants will begin by defining the genetic basis and hallmark clinical features of these syndromes. For instance, TSC is characterized by mutations in TSC1 or TSC2 genes and manifests with cortical tubers, facial angiofibromas, and subependymal giant cell astrocytomas (SEGAs). Neurofibromatosis involves cutaneous neurofibromas, café-au-lait spots, optic gliomas, and peripheral nerve sheath tumors. SWS is associated with facial port-wine stains, leptomeningeal angiomas, and glaucoma, stemming from a somatic mutation in the GNAQ gene.

The course emphasizes applying diagnostic criteria using both clinical evaluation and genetic testing, guiding healthcare professionals in early recognition and accurate classification. Imaging modalities such as MRI, CT, and skin biopsy interpretation are discussed in detail.

Multidisciplinary management strategies will be covered, including neurologic monitoring for seizures and developmental delays, dermatologic interventions such as laser therapy for angiofibromas or port-wine stains, and surgical or pharmacologic interventions for tumor-related complications. Participants will explore the role of mTOR inhibitors like everolimus in TSC, targeted therapies in NF2, and anticonvulsants in SWS.

Emerging innovations such as gene-based therapies, advanced neuroimaging, and molecular diagnostics are integrated into this course to align with future-ready medical practice. By course completion, clinicians will be well-prepared to provide holistic care for patients with neurocutaneous disorders through coordinated, evidence-informed interventions.
