OVERVIEW

Central nervous system (CNS) tumors represent a significant cause of morbidity in pediatric populations, with low-grade gliomas (LGGs) accounting for the most common brain tumor subtype in children. This CME/CE course offers a focused, in-depth exploration of pediatric low-grade gliomas—from clinical presentation and epidemiology to advances in precision medicine and targeted therapeutic strategies.

The course begins with an overview of pediatric LGGs, examining their epidemiological patterns, histological subtypes (such as pilocytic astrocytoma and diffuse astrocytoma), and typical clinical symptoms like headaches, visual disturbances, seizures, or endocrine dysfunction. Participants will gain an understanding of how tumor location and histopathology influence neurologic function and disease trajectory.

Participants will then examine the molecular biology underpinning LGGs, including alterations in genes like BRAF (BRAF-KIAA1549 fusion, BRAF V600E mutation), FGFR1, and MYB/MYBL1. These mutations not only provide diagnostic clarity but also offer therapeutic targets that have transformed pediatric neuro-oncology.

A central focus of this course is the application of precision medicine. Learners will explore how molecular profiling, next-generation sequencing, and RNA expression analysis are integrated into standard-of-care diagnostics to guide individualized treatment plans. Case examples illustrate how therapies targeting BRAF or MEK pathways (e.g., dabrafenib, trametinib) are improving outcomes in refractory or progressive LGGs.

The course also discusses the challenges of implementing precision medicine in real-world pediatric settings—ranging from access to genomic testing to understanding long-term impacts of targeted therapy. Ethical considerations, disparities in care, and integration within multidisciplinary teams are highlighted to promote equitable, evidence-based care.

Upon completion, clinicians will be better prepared to deliver patient-centered, molecularly informed treatment for pediatric patients with CNS tumors, ultimately improving prognosis and quality of life.
