PI3K and PTEN at the Interface of Cell Growth, Vesicular Trafficking and Disease (E1) is organized by Keystone Symposia on Molecular and Cellular Biology and will be held from May 03 - 07, 2020 at Herrenhausen Palace, Hannover, Lower Saxony, Germany. Description This meeting will bring together three decades of research and development in the PI3K/PTEN field and its progression towards the understanding and treatment of human disease. The discovery of rare human disorders due to PI3K/PTEN pathway deregulation is providing important insights into the organismal biology of this pathway and a fertile ground for repurposing of PI3K pathway inhibitors developed as anti-cancer agents. Improved strategies to effectively use PI3K inhibitors in cancer are currently progressing through the clinic, with exciting new opportunities to use PI3K inhibitors in cancer immunotherapy. Ample attention will be given to the mechanism of action of the phosphoinositide lipids, their turnover and their downstream effectors, especially in relation to the class II and III PI3K isoforms whose cellular and organismal functions are slowly being uncovered. Special attention will also be given to the emerging evidence for a critical role for the PI3K/PTEN pathway in neuronal biology. This meeting will bring together basic scientists, drug developers and clinicians, and aims to foster further integration of efforts to ultimately benefit mankind. Description: Among the three major structural biology techniques, NMR spectroscopy bases its strength on the ability of detecting pathways of conformational and dynamic linkages, which are essential for function. Thus, NMR is unique in its power to explain the functional mechanisms of biomolecules and complexes at atomic resolution. In recent years, NMR methodological and technological advances, as well as developments in isotope labeling, have brought high molecular weight complexes into reach of solution-state NMR spectroscopy. In parallel, sophisticated relaxation experiments have allowed the discovery of minor populated states as essential players in the mechanism of function. Last but not least, solid-state NMR spectroscopy has grown to a point where structural and dynamic studies display the same level of resolution and precision as in solution-state NMR, with the advantage of being applicable to molecules of any size. With these capabilities, NMR plays a central role in the description of molecular functions at atomic resolution. In this event, we will review the contribution of NMR spectroscopy to the elucidation of biological mechanisms; in addition we will discuss the future of the field, in particular with respect to the integration of NMR spectroscopy in hybrid structural biology approaches.
