Following the highly successful inaugural Fusion DNA Damage Response for Cancer Therapeutics meeting, the second meeting in this series will take place in July 2027.
Genomic instability and an increased propensity to accumulate DNA damage are defining features of many cancers. Historical cancer therapies, including radiotherapy and DNA-damaging chemotherapy, have exploited this vulnerability, but their clinical benefit is often limited by collateral damage to normal tissues and associated toxicities. Targeted therapeutic approaches based on inhibiting the DNA damage response (DDR) offer the potential to widen the therapeutic window by tailoring treatment to patients whose tumours have specific DDR defects, dependencies or vulnerabilities. DDR biology in cancer differs from that of normal tissue in several therapeutically important ways. These include the loss or attenuation of one or more DDR pathways, which can increase sensitivity to DNA-damaging agents; elevated replication stress; altered inflammatory and immune-priming potential; and the emergence of specific DDR dependencies that may confer sensitivity to single-agent DDR inhibitors. The clinical activity of poly(ADP-ribose) polymerase (PARP) inhibitors in homologous recombination repair-deficient tumours, including BRCA-mutant cancers, remains the exemplar of this concept and continues to provide a foundation for the development of next-generation DDR therapeutic strategies.
This meeting will focus on current and emerging approaches to targeting the DDR for cancer therapy. Key themes will include advances in PARP inhibitor development, DDR therapeutics based on the concept of synthetic lethality, exploiting replication stress and chromosomal instability through checkpoint inhibition, new modalities in DDR drug discovery, biomarker development for patient stratification and enhancing the efficacy of targeted DNA- damaging agents such as antibody-drug conjugates and radioligand therapies. The meeting aims to bring together academic researchers, translational scientists, clinicians and drug developers from academia, biotech, pharma and healthcare. Through scientific presentations, translational case studies and panel discussions, the meeting will provide an opportunity to review progress in the field, critically assess current challenges and identify future directions for DDR-based cancer therapeutics.
