Course Description:
RNA 3' end formation, both pre-mRNA cleavage/polyadenylation and transcriptional termination, constitutes a major control point in eukaryotic gene regulation. The high complexity of eukaryotic transcriptomes, with most genomic space transcribed into either protein coding or long non-coding (lnc) RNA, necessitates tight control of each separate transcription unit (TU). Uncontrolled termination results in downstream gene deregulation as seen in some cancers, cellular stress and viral infection. Regulated RNA 3' end formation (alternative polyadenylation) plays an extensive role in gene regulation by generating mRNAs with different sized 3' UTRs or terminal exons, as well as by promoting premature termination to restrict the expression of unwanted transcripts.
